The PPSQ series of instruments, using automatic Edman degradation of proteins and peptides in combination with HPLC separation and highly sensitive detection of reaction products, as well as powerful software for analysis of results, provide fast and reliable determination of amino acid sequences. The models differ in the number of reactors – 1 reactor (PPSQ-51A) or 3 reactors (PPSQ-53A).

Protein sequencers for laboratories

Working principles
Protein sequencer – equipment for determining the sequence of amino acids from the N-terminus to 50-60 amino acid residues. The basis of amino acid sequence determination is the Edman method – one of the early methods for determining the primary sequence (sequencing) of peptides. The essence of the method is to treat the peptide under study with a certain set of reagents, which leads to the cleavage of one amino acid from the N-terminus of the sequence in the form of its phenylthiohydantoin derivative (Edman reaction). Identification of phenylthiohydantoin derivatives is performed using liquid chromatography under pressure.
Despite the disadvantages of this method (high labor costs and time) and the development of mass spectrometry, it is still used due to the stability and unambiguousness of the results obtained.
Features
- High baseline stability – an isocratic mode of chromatographic separation of phenylthiohydantoin (PTH) derivatives of amino acids is implemented, which achieves baseline stability and high sensitivity of the analysis.
- The isocratic mode of chromatographic analysis of sequencing products provides greater stability of retention time. This allows subtraction of peaks obtained during the analysis of previous cycles, which facilitates the user’s procedure for identifying amino acid sequences.
- Reduced mobile phase consumption and reduced operating costs – the isocratic mode of chromatographic analysis allows for recirculation of the eluent, which significantly reduces mobile phase consumption and reduces the cost of analysis.
- Specialized software for PPSQ series instruments combines the functions of reactor control and liquid chromatograph, which simplifies the analysis of peptides and proteins.
- Additionally, the program allows you to process chromatograms, compare them with each other, and subtract one chromatogram from another to quickly assess the similarities and differences of the analyzed samples.
- Compliance with FDA 21 CFR Part 11 requirements.
